bioWORLD’s Serum Glycoproteome Analysis Kit allows isolation of the serum glycoproteome by M-LAC coupled with LC-MS/MS and the use of GO associations facilitates characterization of a large panel of candidate markers, which can then be evaluated in a particular patient population.
The most abundant proteins present in the serum glycoprotein fraction included a-2-macroglobulin, serotransferrin, haptoglobin, hemopexin, a1-antitrypsin, complement C3, apolipoprotein A-I, a-1-acid glycoprotein, ceruloplasmin, and complement factor H. Using multilectin column consisting of agarose-bound concanavalin A, wheat germ agglutinin, and Jacalin with diluted 100-mL serum equilibrium with GlycoPure™Binding Buffer and eluted with GlycoElute™Elution Buffer, more serum glycoproteins can be captured. After a 15-20 min incubation, the unbound proteins are eluted with 10 mL of equilibrium buffer, and the captured proteins are released with 10mL of a specific GlycoElute™solution.
